MPWG: Basket Trials Sub-stream

Developing confirmatory basket trial methodology and pioneering its application in oncology and rare diseases.

Part of the ASA-DahShu Master Protocol Working Group.

Overview

The Basket Trials sub-stream focuses on the design, simulation, and real-world application of confirmatory basket trials, studies that evaluate a single treatment across multiple diseases or molecular subtypes simultaneously.

Lead: Robert A. Beckman, M.D. (Georgetown University)

Members: Martin Posch (Med U Vienna), Olivier Collignon (GSK), Thomas Gwise (ex-FDA), Nicole Li (BeiGene), Kit Roes (EMA), Kristine Broglio (AstraZeneca), Cindy Lu (AstraZeneca), Philip He (Daiichi Sankyo), Heng Zhou (Merck), Helen Zhou (GSK), Cong Chen (Merck), Linchen He (Novartis), Valeriy Korostyshevskiy (Georgetown), Sathvik Narayana (Albany Medical College), Sammi Tang (Astellas), Ruixiao Lu (Alumis), Patrick Roney (Georgetown)

Focus Areas

  • Educating the scientific community on basket trial methodology, benefits, and appropriate use
  • Developing confirmatory basket trial designs with rigorous Type I error control
  • Simulation-based evaluation of operating characteristics across disease settings
  • Real-world data applications: using external data to inform basket trial design and analysis
